В то же время некоторые из используемых для ее определения методов имеют ряд существенных недостатков и ограничений, и в настоящее время ведется разработка новых более эффективных способов. Целью данной работы явилась сравнительная оценка эффективности применения в клинической лабораторной диагностике двух методов определения активности общей и панкреатической α-амилазы в крови человека: основанного на использовании в качестве субстрата 4,6-этилиден-(G7)-п-нитрофенил-(G1)-α,D-мальтогептаозида (EPS-G7) и 2-хлоро-4-нитрофенил-4-O-β-D-галактопиранозилмальтозида (GalG2CNP). Для этого в параллельных исследованиях проанализированы образцы сыворотки крови пациентов с различной активностью α-амилазы. С использованием очищенных препаратов α-амилазы определены основные аналитические характеристики метода с субстратом GalG2CNP. Полученные результаты продемонстрировали высокий уровень корреляции обоих методов и точности измерения в рабочей области определения активности изоферментов α-амилазы. Таким образом, метод определения активности α-амилазы с использованием в качестве субстрата GalG2CNP обладает аналогичными по сравнению с распространенным методом с субстратом EPS-G7 аналитическими характеристиками, но вместе с тем имеющиеся преимущества позволяют рекомендовать его к более широкому применению в клинической лабораторной диагностике и научных исследованиях.</p></abstract><trans-abstract xml:lang="en"><p>Blood α-amylase activity is a key laboratory marker of pancreatic diseases. At the same time, some of the common laboratory methods of its measurement have a number of limitations. Therefore new more effective laboratory methods are currently being developed over those methods that have been already available. The aim of this work was to compare two methods of determination of the activity of total and pancreatic α-amylase in human blood based on using 4,6-ethylidene-(G7)-p-nitrophenyl-(G1)-α,D-maltoheptaoside (EPS-G7) and 2-chloro-4-nitrophenyl-4-O-β-D-galactopyranosylmaltoside (GalG2CNP) as substrates. Blood serum samples from patients with different levels of α-amylase activity were analyzed. The main analytical characteristics of the method with the GalG2CNP substrate were determined using purified α-amylase specimens. A high-sized correlation and high accuracy of the α-amylase isoenzymes activity were observed for the both methods.
