Цианобактерии как источник питательных и биологически активных соединений представляют собой альтернативный способ лечения. Целью исследования стала оценка терапевтического потенциала внутрижелудочного введения суспензии или экстракта сырой биомассы цианобактерии Leptolyngbya cf. ectocarpi при токсических гепатитах и воспалительном заболевании кишечника у мышей.Материал и методы. Алкогольный, парацетамоловый гепатиты и воспалительное заболевание кишечника у мышей индуцировали внутрижелудочно этанолом, парацетамолом или декстрансульфатом натрия соответственно. Лечение проводили суспензией или экстрактом цианобактерии (внутрижелудочно) за 7 суток до индукции патологического процесса либо через 30 мин после нее; мышам с воспалительным заболеванием кишечника дополнительно назначали циклофосфан или преднизолон. Оценивали количество лейкоцитов, активность печеночных аминотрансфераз, содержание билирубина в крови. Проводили патоморфологическое исследование и морфометрию образцов печени, тонкой и толстой кишки.Результаты и их обсуждение. Введение суспензии и экстракта цианобактерии мышам с алкогольным гепатитом и воспалительным заболеванием кишечника приводило к снижению количества лейкоцитов крови по сравнению с соответствующим контролем, при индукции повреждения печени парацетамолом – к повышению. Показан гепатопротективный эффект цианобактерий при индукции токсических гепатитов (этанол, парацетамол) – уменьшение активности печеночных аминотрансфераз и концентрации билирубина. При токсических гепатитах и воспалительном заболевании кишечника назначение суспензии и экстракта цианобактерии сопровождалось изменениями микроархитектоники печени, тонкой и толстой кишки, которые затрагивали объемные плотности гепатоцитов и синусоидов, количество двуядерных гепатоцитов и синусоидных клеток; глубину крипт, длину ворсинок, толщину слизистой и мышечной оболочки, количество клеток Панета, бокаловидных и интраэпителиальных клеток в тонкой кишке; глубину крипт, толщину подслизистой основы и мышечной оболочки толстой кишки.Заключение. Цианобактерия Leptolyngbya cf. ectocarpi проявляет противовоспалительное и гепатопротектроное действие при гепатитах и воспалительном заболевании кишечника в экспериментальных моделях на мышах.</p></abstract><trans-abstract xml:lang="en"><p>Toxic hepatitis and inflammatory bowel disease are still a medical and social problem in many countries around the world due to their rising incidence, lack of effective treatment options, and need for new therapeutic approaches. Cyanobacteria as a source of nutrients and bioactive compounds represent an alternative treatment method. The aim of the study was to assess the therapeutic potential of intragastric administration of a suspension or extract of raw biomass of the cyanobacterium Leptolyngbya cf. ectocarpi in mice with toxic hepatitis and inflammatory bowel disease.Material and methods. Alcoholic and acetaminophen-induced hepatitis, as well as inflammatory bowel disease, were induced in mice intragastrically with ethanol, acetaminophen, or sodium dextran sulfate, respectively. Treatment was performed with a suspension or extract of the cyanobacterium (given intragastrically) for 7 days before the induction of the pathological process or 30 minutes after it; mice with inflammatory bowel disease were additionally administered cyclophosphamide or prednisolone. Blood bilirubin content, liver aminotransferase activity, and leukocyte number were assessed. Pathomorphological examination and morphometry of liver, small intestine, and large intestine samples were performed.Results and discussion. Administration of the cyanobacterium suspension and extract to mice with alcoholic hepatitis and inflammatory bowel disease led to a decrease in blood leukocyte count compared to the respective control, whereas in the case of acetaminophen-induced liver injury, it led to an increase. When ethanol and paracetamol are used to induce toxic hepatitis, cyanobacteria have been shown to have a hepatoprotective impact by lowering bilirubin content and liver aminotransferase activity. Correction with cyanobacterium suspension and extract revealed alterations in the microarchitecture of the liver, small intestine, and large intestine in cases of inflammatory bowel disease and toxic hepatitis, which affected hepatocyte and sinusoidal volume density, the quantity of binucleated hepatocytes and sinusoidal cells, the depth of crypts, the length of villi, the thickness of the mucous membrane and muscularis, the number of Paneth cells, goblet cells, and intraepithelial cells in the small intestine, and the depth of crypts, the thickness of the submucosal base, and the muscularis in the large intestine.Conclusions.
