Препарат амантадин (АМД, 1-аминоадамантан), известный как блокатор глутаматных NMDA-рецепторов и регулятор нейротрансмиссии в проводниках ЦНС, снижает двигательные расстройства, характерные для БП, однако имеет недостаточную эффективность. В качестве перспективных аналогов аминоадамантанов, сохраняющих способность к стимуляции дофаминергической системы и антагонизма в отношении глутаматной системы, нами предлагаются азаадамантаны, содержащие монотерпеновый фрагмент в боковом положении азаадамантановой структуры.</p><p>Цель исследования – сравнить влияние монотерпеновых производных азаадамантанов (К-372, К-818) и АМД на ключевые нейромедиаторные системы, вовлеченные в патогенез БП, с использованием валидированных моделей in vivo.</p><sec><title>Материал и методы</title><p>Материал и методы. Исследование выполнено на самцах мышей CD-1 и C57BL/6, которым однократно внутрибрюшинно вводили азаадамантаны и АМД в дозе 20 мг/кг. Для оценки нейромедиаторных эффектов использовали модели: индуцированный 1-метил-4-фенил-1,2,3,6-тетрагидропиридином (МФТП) паркинсонизм, галоперидоловую каталепсию, ареколиновый тремор, йохимбиновую токсичность и коразоловые судороги.</p><p>Результаты и их обсуждение. В МФТП-модели К-372 и К-818 достоверно (p &lt; 0,05) улучшали двигательную и ориентировочно-исследовательскую активность, что свидетельствует об их способности стимулировать дофаминергические нейроны; АМД значимо улучшал лишь один показатель двигательной активности. В модели галоперидоловой каталепсии значимую активность наравне с АМД показал только азаадамантан К-818, тогда как К-372 не имел эффекта. В ареколиновой модели К-372 и К-818 увеличили латентный период и уменьшили длительность тремора, а значит, проявили умеренный центральный М-холинолитический эффект, в отличие от АМД, действие которого не было статистически значимым. В модели йохимбиновой токсичности отмечено снижение выживаемости животных, получавших К-372 и К-818, что интерпретируется как потенцирование адренергической передачи; АМД несколько уменьшил токсичность йохимбина. В модели коразоловых судорог не выявлено изменений латентного периода, следовательно, ГАМК-ергическая активность отсутствует у всех агентов, что поддерживает предположение о схожем механизме действия АМД и азаадамантанов.</p></sec><sec><title>Заключение</title><p>Заключение. К-372 и К-818 проявляют выраженную противопаркинсоническую активность в МФТП-модели, а также эффективны в модели паркинсонического тремора у мышей. У агентов обнаружены эффекты потенциации адренергической системы и отсутствие признаков ГАМК-ергической стимуляции.</p></sec></abstract><trans-abstract xml:lang="en"><p>Parkinson’s disease (PD) involves degeneration of nigrostriatal dopaminergic neurons with a consequent imbalance across multiple neurotransmitter systems regulating basal ganglia function. Amantadine (AMD; 1-aminoadamantane) is a low-affinity, uncompetitive NMDA receptor antagonist and a modulator of CNS neurotransmission that alleviates motor symptoms of PD but is limited by modest efficacy. Azaadamantanes bearing a monoterpene moiety are proposed as aminoadamantane analogs with potential to stimulate dopaminergic transmission and antagonize glutamatergic NMDA receptors.</p><p>Aim of the study was to compare the effects of monoterpene-containing azaadamantanes (K-372, K-818) and AMD on key neurotransmitter systems implicated in PD pathogenesis using validated in vivo models.</p><sec><title>Material and methods</title><p>Material and methods. Male CD-1 and C57BL/6 mice received a single intraperitoneal dose of each agent at 20 mg/kg. Outcomes were measured in the following models: parkinsonism, induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP), haloperidol-induced catalepsy, arecoline-induced tremor, yohimbine toxicity, and corazole-induced seizures models.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. In the MPTP model, K-372 and K-818 significantly improved locomotor and exploratory activity, indicating dopaminergic stimulation, whereas AMD significantly improved only one locomotor endpoint (p&lt;0,05). In haloperidol-induced catalepsy, only K-818 showed significant anticataleptic activity comparable to AMD, whereas K-372 had no effect. In the arecoline tremor assay, K-372 and K-818 prolonged latency and reduced tremor duration more effectively than AMD, suggesting a moderate central antimuscarinic action, while AMD produced no significant effect. In yohimbine toxicity model, K-372 and K-818 decreased animal survival, corresponds to potentiation of adrenergic transmission, whereas AMD showed a modest trend toward lower toxicity. In the corazole model, none of the agents altered seizure latency, indicating no evidence of GABAergic stimulation, which aligns with a mechanism resembling amantadine.</p></sec><sec><title>Conclusions</title><p>Conclusions.
