Для решения этих вопросов используют экспериментальные подходы, когда лабораторных животных содержат на высококалорийном рационе длительное время, что позволяет получить модель алиментарного ожирения.</p><p>   Целью работы было провести сравнительный динамический анализ формирования ожирения у крыс, содержавшихся в течение 4 и 7 недель на высокожировой диете (ВЖД).</p><sec><title>   Материал и методы</title><p>   Материал и методы. В работе использовали половозрелых самцов крыс Вистар. Контрольных животных содержали на диете, в которой жир составлял 10–11 %, опытных – на ВЖД, в которой жир составлял 36 % калорийности суточного рациона за счет добавления к пище топленого свиного сала. Измеряли массу органов и жировой ткани, содержание триглицеридов, общего и холестерина липопротеидов высокой и низкой плотности, активности АлАТ и АсАТ в сыворотке крови. Для оценки относительной численности клеток Купфера срезы печени окрашивали гематоксилином и эозином. Для иммуногистохимического исследования срезов применяли непрямой стрептавидин-пероксидазный метод.</p><p>   Результаты и их обсуждение. У крыс, содержавшихся на ВЖД в течение 4 недель, увеличились по сравнению с контрольными животными масса тела (на 21 %), массовый индекс забрюшинного, эпидимального и межлопаточного бурого жира (на 56, 33 и 52 % соответственно), содержание триглицеридов в сыворотке (в 2,3 раза), численная плотность клеток Купфера (в 2,1 раза), экспрессия матриксной металлопротеиназы 9 в печени (в 2,8 раза). Через 7 недель ВЖД активность АлАТ в сыворотке повысилась на 20 %, численная плотность клеток Купфера сохранилась увеличенной в 2,1 раза, уровень экспрессии матриксной металлопротеиназы 9 вырос в 5 раз относительно контрольных крыс.</p></sec><sec><title>   Заключение</title><p>   Заключение. Через 4 недели содержания на ВЖД у крыс сформировался фенотип ожирения, сопровождающийся негативными сдвигами структурно-функционального состояния печени; через 7 недель эти негативные изменения усугубились, что позволяет рассматривать использованную модель ожирения у крыс как инструмент для изучения эффективности подходов к нормализации ожирения и сопутствующих заболеваний.</p></sec></abstract><trans-abstract xml:lang="en"><p>   The aim of this study was to conduct a comparative analysis of the development of obesity in rats fed a high-fat diet (HDL) for 4 and 7 weeks.</p><sec><title>   Material and methods</title><p>   Material and methods. Sexually mature male Wistar rats were used in the work. Control animals were kept on a diet in which fat was 10–11 %, experimental rats – on an HDL, in which fat accounted for 36 % of the caloric content of the daily diet due to the addition of melted pork fat to the food. The mass of organs and adipose tissue, the content of triglycerides, total and cholesterol, high- and low-density lipoproteins, and the activity of alanine and aspartate amino transferase in blood serum were measured. Liver sections were stained with hematoxylin and eosin to assess the relative number of Kupffer cells. An indirect streptavidin peroxidase method was used for immunohistochemical examination of sections.</p><p>   Results and discussion. In rats kept on HDL for 4 weeks, the following parameters increased in comparison with control animals: body weight by 21 %, mass index of retroperitoneal, epidermal and interscapular brown fat (by 56, 33 and 52 %, respectively), serum triglyceride content by 2.3 times, numerical density of Kupffer cells by 2.1 times, the expression of matrix metalloproteinase 9 in the liver by 2.8 times.
